摘要: DNA synthesis techniques and technologies are quickly becoming a cornerstone of modern molecular biology play pivotal role in the field synthetic biology. The ability to synthesize whole genes, novel genetic pathways, even entire genomes is no longer dream it was 30 years ago. Using little more than thermocycler, commercially synthesized oligonucleotides, polymerases, standard laboratory can several kilobase pairs week using existing techniques. Herein, we review used generation DNA, from chemical oligonucleotides their assembly into long, custom sequences. Software websites facilitate execution these approaches explored, applications gene expression discussed. Finally, an example automated our own provided.
