摘要: Abstract Monolayer cultures of the human epidermoid cancer cell line HEp 2 were infected with HF strain herpes simplex virus (herpes hominis) by using an inoculum approximately 1 infectious unit per cell. At 1, 2, 4, and 7 days thereafter studied for number remaining cells, infectivity cells culture medium, cytopathic changes, viral antigen (fluorescent antibody technique) morphology development (electron microscopy). day (16 hours) medium had decreased from initial level about 1% infected. There was no obvious change in only evidence damage a probable reduction cytoplasmic RNA (as judged acridine orange staining) margination chromatin few nuclei. A very sharply concentrated at nuclear membrane or distributed irregularly diffusely throughout nucleus. Rarely contained scattered intranuclear single-membraned particles dense resembling nucleoids membrane. 15% infective there concordant increase DNA, but still major destruction. By 4 destruction apparent on routine microscopy progression process also evident fluorescent electron microscopic studies, characteristic changes infection most frequent study when majority which remained glass Intranuclear inclusions not seen either optical microscopy. On this usually it occurred distinct outlined particles. usual particle consisted spherical nucleoid maximum diameter 60 mμ, surrounded single 100 mμ diameter, empty naked numerous, double-membraned 160 occasionally nucleus advanced infections. Virus never found crystalline arrays. They numerous near contact inner lamella between outer lamellae. striking thickening progressive evagination points particles, seems clear these morphologic alterations that altered becomes second particle. Extreme proliferation plication common, such perinuclear region became complex mixture membranous projections. This produced structures pods peas, enclosing row single- cytoplasm almost always surrounded, individually group, thin third membrane, thus resulting triple total 185 to 200 cluster double thin-walled sac. results are compared previously published studies other viruses group.
