Advances in isolation and characterization of homogeneous cell populations using laser microdissection

摘要: Summary. The isolation and characterization of homogeneous cell populations are great importance for the analysis gene expression, because normal tissues contain various types cells, differences in isolated cells exert significant effects on expression analysis. Researchers have attempted to develop methods populations, such as flow cytometry mechanical dissection. However, recent emergence laser-assisted microdissection has revolutionized single-cell from solid tissues. With help a cutting laser, laser can isolate (cells) interest without contamination surrounding with microscopic visualization field. By combining subsequent microarray technology, several studies resulted identification disease-related genes. In this review, we summarize principle provide successful examples target-gene using conventional method microarray. Next, discuss practical drawbacks combinational method, need large number disturbance relative abundance transcripts during RNA amplification. We introduce our modifications combined detection genes; technique is simple, yet accurate. Finally, versatile applications microdissection, not only transcript analysis, but also other genomics proteomics analyses are, presented. Histol Histopathol 20, 139-146 (2005)
