Selection procedure for deregulated iron transport mutants (fur) in Escherichia coli K 12: fur not only affects iron metabolism

摘要: A selection procedure using Mn2+ is described. high percentage of the resistant mutants had constitutive iron transport systems. By P1 transduction, and complementation with cloned fur gene it could be shown that nearly all in expression operon fusion fiu::lambda placMu were only defective fur. High concentrations manganese inhibited derepression an iron-regulated lac fusion. In another was inducible by iron, also induced production beta-galactosidase. Most isolated (80%) not able to grow on succinate, fumarate or acetate. After transformation a fur+ plasmid 39 tested succinate. presence succinate growth medium reduced uptake rates 50%-70%. Succinate dehydrogenase activity 10% parent strain.
