摘要: The induction and establishment of hairy root cultures Gentiana dinarica using two strains Agrobacterium rhizogenes (A4M70GUS 15834/PI) is reported for the first time. Strain 15834/PI had higher rate roots (32.15 %) than strain A4M70GUS (6.12 %). Transgenic nature was confirmed by GUS assay PCR analysis. Two clones per (A4M70GUS-D -I, 15834/PI-2 -3) with marked differences in general morphology growth were further studied. methanol extracts analyzed xanthones content HPLC method. effects type carbohydrate source (sucrose, fructose or glucose) at different concentrations on parameters (growth index, dry weight, fresh/dry weight ratio), phenolic xanthone production, free radical scavenging activity transgenic evaluated. Statistical level factorial design used to define optimal conditions successful secondary metabolite production G. clones. results showed that A4M70GUS-D 15834/PI-3 superior ones. These highest 116.8 mM sucrose, producing up threefold amounts total phenolics norswertianin-1-O-primeveroside other clones, untransformed wild-grown plants.
