摘要: Regulation of albumin gene expression is believed to be mediated by multiple nuclear factors that interact with cis-acting DNA sequences within the first 160 base pairs (bp) promoter. The minimal promoter sequence required generate tissue-specific has not been clearly defined. We have constructed a series transient vectors containing progressive deletions mouse 5′-flanking fused bacterial chloramphenicol acetyltransferase (CAT) and include Moloney murine leukemia viral (Mo-MuLV) enhancer. Promoter activity was determined in hepatoma fibroblast cell lines S1 nuclease analyses. All constructions were compared −623 Albcat-Mo-MuLV which contains all homology between rat promoters. Low levels observed −60 (10%) but cells. Addition −208 bp progressively increased 190% cells, while −308 −1612 had similar level, −3000 showed 2-fold reduction transcriptional activity. inclusion upstream generated low fibroblasts. also show from liver extracts protect at least five regions −160. Our results indicate tissue specificity established proximal region additional elements may functional role efficiency are located −160 bp.
