Effects of a miR-31, Runx2, and Satb2 Regulatory Loop on the Osteogenic Differentiation of Bone Mesenchymal Stem Cells

摘要: Recently, a cohort of miRNAs, including miR-31, was reported to be downregulated during osteogenic induction by miR microarray analysis. It remains unclear how changes in miR-31 expression collaborate with bone transcription factors activate the biological pathways that regulate differentiation mesenchymal stem cells (BMSCs). Here effects Runx2, and Satb2 on BMSCs were investigated using mimics inhibitors small interfering RNA for knockdown Runx2 plasmids overexpression Runx2. Our results showed decreased progressively BMSC cultures differentiation. Inhibition dramatically increased alkaline phosphatase activity mineralization cultures. Additionally, diminished levels protein without significantly affecting mRNA levels, directly repressed expression. Overexpression reduced OPN, BSP, OSX, OCN, but not Furthermore, high cultured proliferation medium which may contribute maintenance an undifferentiated state. In conclusion, our suggest Satb2, regulatory mechanism play important role inducing The this study provide us better understanding molecular mechanisms govern fate.
