Differential activity of the promoter for the human alcohol dehydrogenase (retinol dehydrogenase) gene ADH3 in neural tube of transgenic mouse embryos.

摘要: Mammalian alcohol dehydrogenase (ADH) has previously been shown to function in vitro as a retinol well an ethanol dehydrogenase. Thus ADH participates the conversion of (vitamin A alcohol) retinoic acid, regulatory ligand for acid receptor class transcription factors. Human exists family isozymes encoded by seven genes, which are differentially expressed adult liver and extrahepatic tissues, being found preferentially epithelial cells retinoid target tissues. However, human expression patterns have not analyzed early embryonic known synthesize respond such neural tube limb buds. To estimate pattern one member family, we constructed transgenic mouse lines carrying ADH3 promoter fused lacZ gene. ADH3-lacZ transgene was first noted at day 9.5 active extending from midbrain spinal cord, heart proximal regions forelimb In 12.5 13.5 embryos, remained also observed more distal hindlimb buds kidney. Expression highest ventral midline including floor plate showed dorsal gradient decreasing expression. These findings indicate that least isozyme may exist correct tissues act catalyzing synthesis acid.
