摘要: Thermodynamics of 39 oligonucleotides with internal G·T mismatches dissolved in 1 M NaCl were determined from UV absorbance versus temperature profiles. These data combined literature values six sequences to derive parameters for 10 linearly independent trimer and tetramer Watson−Crick base pairs. The mismatch predict ΔG°37, ΔH°, ΔS°, TM average deviations 5.1%, 7.5%, 8.0%, 1.4 °C, respectively. predictions are within the limits what can be expected a nearest-neighbor model. show that contribution single helix stability is context dependent ranges +1.05 kcal/mol AGA/TTT −1.05 CGC/GTG.
