摘要: Seven DNA extraction protocols were used to obtain from herbarium specimens ofJuncus andLuzula (Juncaceae) of various ages. historical samples is difficult extract, and the extracts are seldom good quality. The quality obtained was estimated by using a spectrophotometer measure A260/280 absorbance ratio. total yield measured fluorometer. results indicate success both mixer mill grinding DNeasy Plant Kit. Another protocol (grinding with mortar pestle, liquid nitrogen) yielded many samples. Modified CTAB extraction, lengthy precipitation, usually provided amounts DNA. Other did not give satisfactory results.
