摘要: We have identified a human insulinoma cDNA (PC2) that encodes protein homologous to the precursor processing Kex2 endoprotease of yeast by using polymerase chain reaction detect and amplify conserved sequences within catalytic site. The 638-residue amino acid sequence PC2 begins with cleavable signal peptide, indicating it enters secretory pathway, contains 282-residue domain is modules both related bacterial subtilisins. Within this region 49 27% acids are identical those in aligned subtilisin BPN' sequences, respectively, catalytically essential Asp, His, Ser residues all conserved. Northern blot analysis revealed presence 2.8- 5.0-kilobase hybridizing bands mRNA from insulinoma. also shows great similarity incomplete NH2-terminal furin gene product, putative membrane-inserted receptor-like molecule. propose member family mammalian Kex2/subtilisin-like proteases includes members involved number specific proteolytic events cells, including prohormones.
