Overproduction of inhibitory hematopoietic cytokines by lipopolysaccharide-activated peripheral blood mononuclear cells in patients with aplastic anemia

摘要: The aim of this study was to measure the level cytokines produced by peripheral blood mononuclear cells (PBMNC) in patients with aplastic anemia (AA) and determine their effect on clonal growth normal bone marrow (BM) cells. Twenty-one AA 11 controls were enrolled study. Medium conditioned PBMNC presence lipopolysaccharide (LPS) found be suppressive colony BM Thus, we further determined PBMNC-conditioned medium (CM) both inhibitory cytokines: macrophage inflammatory protein-1α (MIP-1α), tumor necrosis factor-α (TNF-α), transforming factor-β2 (TGF-β2), interferon-γ (IFN-γ), stimulatory interleukin-3 (IL-3) stem cell factor (SCF). Spontaneous production MlP-1α higher than (1887±174 pg/ml vs 1643±93 pg/ml), but difference not significant. After LPS stimulation, MIP-1α markedly increased patients, its significantly that (2360±149 1517±92 pg/ml, p=0.0022). TNFα also patients. However, IFN-γ, TGF-β2, SCF, IL-3 detectable PBMNC-CM either or normals. myelopoietic suppressing AA-PBMNC-CM from each patient blocked pretreatment anti-TNF-α, resulting a colony-forming enhancement 174%±12%.
