Quantitative analysis of human milk oligosaccharides by capillary electrophoresis.

摘要: Human milk oligosaccharides may have important biological activities. 1 We developed a sensitive, convenient, quantitative method for the routine study of sialylated (acidic, negatively charged) in large numbers samples. Capillary electrophoresis (CE) with detection at 205 nm was sensitive to femtomole level and could resolve quantify nine acidic milk, ranging from tri- nonasaccharides. Milk samples Massachusetts, South Carolina, Mexico were analysed Hewlett Packard 3D capillary apparatus (buffer: 376 mM Trizma H [pH 7.91 150 SDS, plus 6% [v/v] methanol as an organic modifier; approximately one nanoliter loaded onto column each analysis). Peaks identified by coelution reference oligosaccharides: 3'-sialyllactose (3'-SL), 6'-sialyllactose (6'-SL), disialyltetraose (DST), 3'-sialyl-3-fucosyllactose (3'-S-3-FL), sialyl1acto-Ntetraose-a (SLNT-a), sialyllacto-N-tetraose-b (SLNT-b), sialyllacto-Nneotetraose-c (SLNT-c), disialyllacto-N-tetraose (DSLNT), disialomonofucosyllacto-N-neohexaose (DSFLNH). The lower limit 20 70 femtomoles. Typical electropherograms pooled contained 16 well-defined peaks eluting between 15 30 min. Seven major coeluted SLNT-b, SLNT-c, SLNT-a, 3'-S-3-FL, 6'-SL, 3'-SL,
