摘要: An integrated approach involving physical mapping, identification of transcribed sequences, and computational analysis genomic sequence was used to generate a detailed transcription map the 1.0-Mb region containing breast cancer susceptibility locus BRCA2 on chromosome 13q12–q13. This is included in genetic interval bounded by D13S1444 D13S310.Retrieved sequences from exon amplification or hybrid selection procedures were grouped into intervals subsequently units clone overlap. Overlap established direct hybridization, cDNA library screening, PCR linking (island hopping), and/or alignment. Extensive sequencing performed an effort understand unit organization. In total, approximately 500 kb completed. The further characterized hybridization RNA series human tissues. Evidence for seven genes, two putative pseudogenes, nine additional obtained. One recently identified as but all others are novel genes unknown function only limited alignment public databases observed. large gene with transcript size 10.7 showed significant similarity predicted Caenorhabditis elegans genome Saccharomyces cerevisiae efforts, while another contained motif similar 2′, 3′ cyclic nucleotide phosphodiesterase gene. Several retrieved not aligned because no corresponding cDNAs obtained when screening libraries lack definitive evidence splicing signals coding based analysis. However, presence suggested groups exons selected clones that consistent orientations could be localized common intervals.
