ZnRF3 Induces Apoptosis of Gastric Cancer Cells by Antagonizing Wnt and Hedgehog Signaling

摘要: A large proportion of malignant cancers the stomach are gastric adenocarcinoma type. In spite many studies, molecular basis for this cancer is still unclear. Deregulated cell proliferative signaling via Wnt/β-catenin and Hedgehog pathways considered important in pathogenesis including cancer. Recent studies identified ZnRF3 protein, which a E3-ubiquitin ligase either deleted or mutated cancers, to inhibit Wnt signaling. However, significance control whether it also regulates pathway, not known. present study, we assessed expression tumors paracancerous tissues from 58 patients (44 male 14 female) different ages related patient survival. We observed clear relationship between tissue tumor size. Also, was much higher aged patients. Male showed mortality than females. Mechanistic using normal cells (GES1) (MGC-803) infected with AdZnRF3 AdGFP viral vectors, revealed that overexpression causes significantly more apoptosis lowered proliferation cells. led greatly reduced levels Lgr5, component Gli1, Thus, negatively influences both pathways, probably way progression. These results suggest importance function checking progression growth indicate lack protein can lead poorer clinical outcomes
