Effect of adjunctive lamotrigine treatment on the plasma concentrations of clozapine, risperidone and olanzapine in patients with schizophrenia or bipolar disorder.

摘要: The effect of lamotrigine on the steady-state plasma concentrations atypical antipsychotics clozapine, olanzapine, and risperidone was investigated in patients with schizophrenia or bipolar disorder stabilized chronic treatment clozapine (200-500 mg/day; n = 11), (3-6 10) olanzapine (10-20 14)). Lamotrigine titrated up to a final dosage 200 mg/day over 8 weeks, pharmacokinetic assessments were made at baseline during weeks 6 10, dosages 100 respectively. norclozapine, risperidone, 9-hydroxy-risperidone did not change significantly lamotrigine. mean 31 +/- 7 ng/mL baseline, 32 week 6, 36 9 difference between 10 being statistically significant (P < 0.05). Adjunctive therapy well tolerated all groups. These findings indicate that lamotrigine, recommended for use as mood stabilizer, does affect levels their active metabolites. modest elevation concentration, possibly due inhibition UGT1A4-mediated glucuronidation, is unlikely be clinical significance.
