Effects of granulosa cell co-culture on in-vitro meiotic resumption of bovine oocytes

摘要: This study was undertaken to create an in-vitro model using granulosa cell monolayers replace the role of follicle in maturation bovine oocytes. Cumulus-oocyte complexes were co-incubated with fresh or 7-day cultures (with new conditioned medium) medium alone, presence absence IBMX (isobutylmethylxanthine), adenosine heparin. Progression metaphase-II stage significantly affected by co-culture oocytes and a lesser degree when cultured supernatant alone (conditioned medium). The attached rapidly monolayer, suggesting that intimate contact between cells cumulus-oocyte is important signal for maintenance meiotic arrest. Heparin did not prevent itself, but prevented attachment monolayers, thereby reducing their inhibitory effect. Adenosine cumulus expansion reduced effective inhibitor only additional cells.
