Gentamicin and leucine inhalable powder: what about antipseudomonal activity and permeation through cystic fibrosis mucus?

摘要: The aim of this study was to evaluate the permeation properties gentamicin (G) in a novel dry powder form for inhalation through an artificial mucus model. Moreover, since respiratory infections sustained by Pseudomonas are major cause sickness and death CF patients, susceptibility P. aeruginosa engineered G powders investigated. Micronized G/leucine (85:15) formulations were produced co-spray-drying, using process parameters conditions previously set. Powders characterized terms yield, drug content aerodynamic profiles, analyzed Andersen Cascade Impactor. Different models prepared, showing composition viscosity similar those native mucus. To investigate impact on permeation, Franz-type vertical diffusion cells used; applied directly synthetic membrane with or without interposition layer. In buffer, showed controlled release; presence leucine reduced wettability and, consequently, rate. Otherwise, delayed from both formulations, faint influence aminoacid. Antimicrobial tests revealed that G/leu particles able preserve antipseudomonal activity, even
