摘要: Single-molecule manipulation has allowed the forced unfolding of multidomain proteins. Here we develop a theory that not only explains these experiments, but also points out number difficulties in their interpretation and makes suggestions for further experiments. Our is valid essentially any molecule can be unfolded AFM: as an example present force-extension curves both titin RNA hairpins. For reproduce curves, dependence break force on pulling speed, break-force distributions, validate two common experimental views: Ig domains explained stepwise increases contour length, increasing peaks native sequences represent hierarchy bond strengths.
