Reduced ribosomal DNA transcription in the prefrontal cortex of suicide victims: consistence of new molecular RT-qPCR findings with previous morphometric data from AgNOR-stained pyramidal neurons.

摘要: Prefrontal cortical regions play a key role in behavioural regulation, which is profoundly disturbed suicide. The study was carried out on frozen samples from the anterior cingulate cortex (dorsal and ventral parts, ACd ACv), orbitofrontal (OFC), dorsolateral (DLC) obtained 20 suicide completers (predominantly violent) with unknown psychiatric diagnosis 21 non-suicidal controls. relative level of ribosomal RNA (rRNA) as marker transcriptional activity DNA (rDNA) evaluated bilaterally prefrontal mentioned above (i.e. eight interest, ROIs) by reverse transcription quantitative polymerase chain reaction (RT-qPCR). overall statistical analysis revealed decrease rDNA victims versus controls, particularly male subjects. Further ROI-specific post hoc analyses significant this suicides compared to non-suicides five ROIs. This effect accentuated ACv, where it observed bilaterally. Our findings suggest that decreased plays an important pathogenesis corresponds our previous morphometric AgNOR-stained neurons.
