摘要: Over the recent year we have witnessed considerable advances in study of neurotransmitter release. This progress has been severalfold as different techniques allowed us to characterise many steps along process exocytosis, membrane fusion, formation fusion pore, and given insight kinetics release re-uptake. Patch clamping provided quantitative measurements capacitance changes secretory vesicle is added surface cell during secretion, change opposite direction when retrieved back into exocytosis. Carbon-fibre microelectrodes measured electrochemically oxidisable transmitters extracellular space. Differential interference contrast microscopy spatially resolved images exocytosis; real-time that are suggestive bubbles breaking a boiling pot water. The interest novel stems from fact existing approaches can provide only indirect evidence on preceding fusion. vesicular dynamics just beneath plasma out reach or amperometric detection. What needed tool would allow look below surface. much-needed appears be evanescent-wave microscopy. review describes how laser used exocytosis at single-vesicle resolution. A companion paper deals with practical aspects imaging.
