NeuCode Labeling in Nematodes: Proteomic and Phosphoproteomic Impact of Ascaroside Treatment in Caenorhabditis elegans

摘要: The nematode Caenorhabditis elegans is an important model organism for biomedical research. We previously described NeuCode stable isotope labeling by amino acids in cell culture (SILAC), a method accurate proteome quantification with potential multiplexing beyond the limits of traditional culture. Here we apply SILAC to profile proteomic and phosphoproteomic response C. two potent members ascaroside family pheromones. By consuming labeled E. coli as part their diet, nematodes quickly easily incorporate heavy lysine isotopologues young adult stage. Using this approach, report, at high confidence, one largest data sets date elegans: 6596 proteins false discovery rate ≤ 1% 6620 phosphorylation isoforms localization probability ≥75%. Our reveal post-translational signature pheromone sensing that includes many conserved implicated longevity stress.
