Activation-induced cytidine deaminase auto-activates and triggers aberrant gene expression.

摘要: DNA methylation is a well-characterized epigenetic landmark involved in transcriptional regulation; however, mechanisms underlying its regulation remain poorly characterized. Recent studies demonstrate that activation-induced cytidine deaminase (AID) active demethylation. AID aberrantly expressed inflammation-associated cancers and generates point mutations; cellular disorders attributed to demethylation function are largely unexplored. Here we ectopic expression perturbs tumor-related gene expression. (with Gadd45) activated methylated paired box 5 (Pax5) reporter construct, induced association of endogenous Pax5 with the promoter, suggesting aberrant triggers an auto-activation circuit consolidate self-expression.
