摘要: Background Osteosarcoma (OS) is a malignant bone tumour that exhibits high mortality. While tumours thrive in state of malnutrition, the mechanism by which OS cells adapt to metabolic stress through reprogramming remains unclear. Methods We analysed expression ROCK2 osteosarcoma tissues RT-qPCR and Western blot. Cell proliferation were using CCK8, EdU colony formation assays. The level cell glycolysis was detected glucose-6 phosphate, glucose consumption, lactate production ATP levels. Results Herein, our study showed higher than adjacent tissues. Functional assays have demonstrated contributes growth inducing aerobic glycolysis. current revealed knockdown decreased levels mitochondrial hexokinase II (HKII). And also indicated served as key enzyme it an important role growth. A significant positive correlation identified between mRNA protein expressions HKII, further demonstrating ROCK2-induced dependent on HKII cells. Mechanistically, promotes activating phospho-PI3K/AKT signalling pathway. Conclusion Taken together, results linked two drivers new control OS.
