摘要: The stability of monosaccharides in methanolic hydrochloric acid different strengths and at temperatures was determined. They are generally stable for 24h 1m- 2m-hydrochloric both 85°C 100°C, but undergo considerable destruction 4m- 6m-hydrochloric 100°C. Analysis glycopeptides oligosaccharides known composition showed that release carbohydrate complete within 3h 1m-hydrochloric 85°C. Removal by rotary evaporation resulted losses monosaccharides, which could be prevented prior neutralization. Methanolysis caused extensive de-N-acetylation acetamidohexoses, so a re-N-acetylation step is necessary the analytical procedure. addition acetic anhydride this purpose also loss internal standard adsorption on insoluble silver salts used Several trimethylsilylating agents were studied suitable conditions recommended. effects system water some common organic inorganic contaminants assessed.
