摘要: Although advances in sequencing technologies have popularized the use of microRNA (miRNA) (miRNA-seq) for quantification miRNA expression, questions remain concerning optimal methodologies analysis and utilization data. The construction a library selects RNA by length rather than type. However, as we previously described, miRNAs represent only subset species obtained size selection. Consequently, libraries also contain variety additional small RNAs. This study looks at prevalence these other from bone marrow aspirate specimens explores predictive value RNAs determination response to therapy myelodysplastic syndromes (MDS). Paired pre post treatment were patients with MDS who treated either azacytidine or decitabine (24 pre-treatment specimens, 23 post-treatment specimens) 22 non-MDS control specimens. Total was extracted submitted next generation after an exclusion step enrich enumerated, single nucleotide variants (SNVs) identified, finally differential expression tRNA-derived (tDRs) correlated diseasestatus therapy. Using data generated samples known (N = 47) controls (N = 23), demonstrated that transfer (tRNA) fragments (specifically tRNA halves, tRHs) are one most common isolated values data, identified six differentially expressed between normal samples. elastic net method, four tRNAs-derived together can explain 67 % variation patients. Similar specifically mitochondrial tDRs (mt-tDRs) 13 mt-tDRs which distinguished disease status mt-tDR predited response. Finally, 14 SNVs within found least 20 % not observed any highlights RNA-seq studies focused on potential etiologies species, both technical biologic, discussed well important challenges interpretation tDR Our results suggest be accurately detected through may useful diagnosis prediction
