Development of potentiometric urea biosensor based on urease immobilized in PVA–PAA composite matrix for estimation of blood urea nitrogen (BUN)

摘要: A urea biosensor was developed using the urease entrapped in polyvinyl alcohol (PVA) and polyacrylamide (PAA) composite polymer membrane. The membrane prepared on cheesecloth support by gamma-irradiation induced free radical polymerization. performance of monitored a flow-through cell, where kept conjugation with ammonia selective electrode added as substrate phosphate buffer medium. produced result enzymatic reaction potentiometrically. potential system amplified an electronic circuit incorporating operational amplifiers. Automated data acquisition carried connecting output to 12-bit analog digital converter card. sensor working range 1-1000 mM response time 120 s. enzyme membranes could be reused 8 times more than 90% accuracy. tested for blood nitrogen (BUN) estimation clinical serum samples. showed good correlation commercial Infinitytrade mark BUN reagent method chemistry autoanalyzer. preserved containing dithiothreitol, beta-mercaptoethanol glycerol period two months without significant loss activity.
