摘要: The free-living soil protozoon Acanthamoeba castellanii synthesizes a range of polyunsaturated fatty acids, the balance which can be altered by environmental changes. We have isolated and functionally characterized in yeast microsomal desaturase from A. castellanii, catalyzes sequential conversion C16 C18 Δ9-monounsaturated acids to di- tri-unsaturated forms. In case substrates, this bifunctional Δ12,Δ15-desaturase generated highly unusual acid, hexadecatrienoic acid (16:3Δ9,12,15(n-1)). identification desaturase, catalyze insertion double bond between terminal two carbons represents new addition functionality plasticity. also co-expressed with Δ6-desaturase, resulting synthesis unsaturated hexadecatetraenoic (16:4Δ6,9,12,15(n-1)), previously only reported marine microorganisms. Our work therefore demonstrates feasibility heterologous n-1 series. presence is considered reference evolution desaturases lineage protist.
