The replication initiator protein of a geminivirus interacts with host monoubiquitination machinery and stimulates transcription of the viral genome

摘要: Geminiviruses constitute a group of plant viruses, with ssDNA genome, whose replication in the nucleus an infected cell requires function geminivirus-encoded initiator protein (Rep). Our results suggest that monoubiquitinated histone 2B (H2B-ub) promotes tri-methylation 3 at lysine 4 (H3-K4me3) on promoter Chilli leaf curl virus (ChiLCV). We isolated homologues two major components monoubiquitination machinery: UBIQUITIN-CONJUGATING ENZYME2 (NbUBC2) and HISTONE MONOUBIQUITINATION1 (NbHUB1) from N. benthamiana. ChiLCV failed to cause disease NbUBC2-, NbHUB1-silenced plants, same time, H2B-ub H3-K4me3 modifications were decreased, occupancy RNA polymerase II viral was reduced as well. In further investigations, Rep found re-localize NbUBC2 cytoplasm nucleoplasm, like NbHUB1, cognate partner NbUBC2. observed interact co-localize NbHUB1 nuclei cells. summary, current study reveals binds genome interacts for subsequently trimethylation mini-chromosomes enhances transcription genes.
