Automated Molecular Pathology : One Hour In Situ DNA Hybridization

摘要: AbstractWe describe a rapid automated technique for colorimetric in situ DNA hybridization using the Code-On Immuno-DNA Slide Stainer. We report recent advances positively charged glass slide technology, limolene/xylene dewaxing, stable ribonuclease and pepsin predigestion, heat cycling during hybridization, alkaline phosphatase enhancement. These amplify signal to noise ratio of molecular point where complete one hour assays can be performed from dewaxing counterstaining by automation. Both biotin labeled oligonucleotide nick translated probes localize their viral targets with this single, mechanized protocol at rate per minute. (The J Histotechnol 14:219, 1991)
