A comparison of the effects of diverse mutagens at the lacZ transgene and Dlb-1 locus in vivo.

摘要: Transgenic assays permit the detection of mutations in any tissue, whereas endogenous can be measured very few. For this reason comparisons between these loci when both same cells are considerable interest. Previous have been inconsistent: usually responded alike, however, some cases locus has more sensitive and at other times transgenic sensitive. Here we report a comparison lacZ transgene MutaMouse Dlb-1 gene epithelium small intestine after acute exposure to seven mutagens. Benzo[a]pyrene, 5-bromo-2'-deoxyuridine, methyl methane sulphonate, ethyl N-ethyl-N-nitrosourea, mitomycin C N-methyl-N-nitrosourea were all given by gavage F1 (MutaMouse x SWR) mice. Mutations quantified 2 weeks end treatment. The data shows that agents induced similar mutant frequencies transgene. treatments generally produced only modest increases frequency loci. higher background observed reduces ability assay detect absolute increase frequency.
