Morphological and biochemical changes during formocresol induced cell death in murine peritoneal macrophages: apoptotic and necrotic features

摘要: The present study was conducted to investigate the role of Formocresol (FC)-induced apoptosis and necrotic cell death in murine peritoneal macrophages (pMo). Macrophages were cultured with 1:100 FC for 2 24 h. viability (trypan blue assay), morphology (scanning electronic microscope), apoptotic indexes (light fluorescent microscopy) determined at different scheduled times. Simultaneously, expressions proteins related stress, survival, measured by western blotting. FC-exposed exhibited maximal from 6 h, coincident Bax overexpression (P < 0.001). Additionally, Bcl-xL showed expression between 12 24 h suggesting its survival effect pMo. lowest pMo increment rate 4 12 h observed accordance Fas Hsp60 overexpressions. In summary, all experimental data suggest that two pathways emerge exposed FC, one leading Bax-dependent (2–6 h) other favoring necrosis (4–18 h), Fas-receptor stress signal.
