摘要: Abstract The filamentous fungi Trichoderma reesei is widely used in the production of cellulolytic enzymes and recombinant proteins. However, only moderate success has been achieved expressing heterologous proteins T. reesei. Light-dependent control DNA transcription, protein expression have demonstrated bacteria, fungi, mammalian cells. In this study, light inducible transactivators, a “light-on” bidirectional promoter “light-off” were constructed successfully for first time. Our transactivators can homodimerize bind to upstream region artificial promoters activate or repress genes transcription. Additionally, we upgraded light-inducible system on-off that simultaneously multiple Moreover, native cellulase-free background was by knocking out involved transcriptional regulation encoding cellulases: xyr1, cbh1, cbh2. light-switchable showed very little robust activation blue with significantly improved expression. We demonstrate our potential application as an on/off “switch” regulate under background.
