The CPP Tat enhances eGFP cell internalization and transepithelial transport by the larval midgut of Bombyx mori (Lepidoptera, Bombycidae)

摘要: Cell-Penetrating Peptides (CPPs) are short peptides that able to translocate across the cell membrane a wide range of cargoes. In past decade, different mammalian lines have been used clarify mechanism CPPs penetration and characterize internalization process, which has described either as an energy-independent direct through plasma membrane, or endocytic uptake. Whatever involved, properties these make their use very attractive vector for promoting cellular uptake coupled bioactive macromolecules, such peptides, proteins oligonucleotides. Here we demonstrate, first time in insect, cultured columnar cells from larval midgut Bombyx mori more readily internalize eGFP (enhanced Green Fluorescent Protein) when fused CPP Tat. Tat-eGFP translocates absorptive non-endocytic manner, since no inhibition fusion protein is exerted by metabolic inhibitors drugs interfere with Moreover, Tat enhances intact tissue, mounted suitable perfusion apparatus, transepithelial flux protein. These results open new perspectives effective delivery insecticidal macromolecules targeting receptors located both within insect gut epithelium behind barrier, hemocoel compartment.
