摘要: Circulating microRNAs (miRNAs) are emerging as promising biomarkers for several disorders and related pain. In equine practice, acute laminitis is a common disease characterised by intense pain that severely compromises horse welfare. Recently, the Horse Grimace Scale (HGS), facial expression-based coding system, was shown to be valid welfare indicator identify linked laminitis. The present study aimed to: determine whether miRNAs can used in horses (Equus caballus) affected laminitis; integrate their target genes categorise biological processes; gather additional evidence on concurrent validity of HGS investigating how it correlates miRNAs. Nine presenting with no prior treatment were recruited. As control group, nine healthy further included experimental design. Samples collected from at admission before any ('pre-treatment') 7 days after routine ('post-treatment'). expression levels circulating miRNAs, namely hsa-miR-532-3p, hsa-miR-219-5p, mmu-miR-134-5p, mmu-miR-124a-3p, hsa-miR-200b-3p, hsa-miR-146a-5p, hsa-miR-23b-3p, hsa-miR-145-5p hsa-miR-181a-5p, detected assessed potential quantitative PCR using TaqMan® probes. area under receiver operating curve (AUC) then evaluate diagnostic performance Molecular data integrated scores one trained time point blind veterinarian. comparative analysis demonstrated miR-23b-3p (P=0.029), miR-145-5p (P=0.015) miR-200b-3p (P=0.023) significantly higher pre-treatment AUCs 0.854, 0.859 0.841, respectively. MiR-200b-3p decreased (P=0.043). Combining two panel, miR-200b-3p, increased efficiency distinguishing animals controls. addition, deregulated positively correlated scores. Computational prediction functional enrichment identified pathways between different particular, glutamatergic pathway all three suggesting crucial role pathogenesis conclusion, dynamic miR-23b-3p, considered potentially biomarkers. Further studies needed order assess relevancy other painful conditions compromising An important implication would possibility use them validation non-invasive indicators horses.
