Development of a novel bead-based 96-well filtration plate competitive immunoassay for the detection of Gentamycin

摘要: We developed a sensitive, simple, inexpensive and rapid bead-based immunoassay platform, composed of liposomal nanovesicle amplification system, Gentamycin sulfate beads 96-well filtration plates. In the beginning assay, beads, specific antibody were incubated in bottom-sealed plate. After incubation, washing was done by running buffer through unsealed plate with only gravity antibody-Gentamycin bead complexes retained Fluorescent dye-loaded protein G-liposomal nanovesicles then added to specifically bind antibodies on beads. unbound nanovesicles, millions fluorescent dye molecules released adding detergent solution lyse nanovesicles. The limit detection (LOD) this novel platform TBS skim milk 52.65 ng/mL 14.16 ng/mL, which are both sufficient for detecting 200 Codex maximum residual level (MRL). dynamic ranges from each their LODs 100 μg/mL. 50% inhibition concentrations (IC50) 199.66 360.81 respectively. also demonstrated good specificity comparing results between pure mixture 6 different antibiotics including milk. entire assay 60 samples conducted within 2h. sum, biosensing not fulfilled most benefits magnetic assays, but convenient replacing separation separation.
