Assessing the genetic diversity in Argopecten nucleus (Bivalvia: Pectinidae), a functional hermaphrodite species with extremely low population density and self‐fertilization: Effect of null alleles

摘要: Argopecten nucleus is a functional hermaphroditic pectinid species that exhibits self-fertilization, whose natural populations have usually very low densities. In the present study, genetic diversity of wild population from Neguanje Bay, Santa Marta (Colombia), was estimated using microsatellite markers, and effect presence null alleles on this estimation assessed. A total 8 markers were developed, first described for species, their amplification conditions standardized. They used to determine genotype 48 individuals Naguanje 1,010 derived offspring 38 directed crosses. For each locus, frequencies identified alleles, including statistical package Micro-Checker, parental genotypes confirmed segregation analysis. Three per locus with 0.001 0.632 detected. The ranged 0.10 0.45, Ho 0.0 0.79, He 0.53 0.80. All loci in H-W disequilibrium. allele values high, lower estimations analysis than Micro-Checker. results show high levels indicate not only cause deviation equilibrium all loci, suggesting under study presents signs inbreeding Wahlund effect.
