摘要: We hypothesized that the regulation of apoptosis is an important determinant capillary network structure. Using human umbilical vein endothelial cells (HUVEC) in vitro model systems tube formation, we initially documented a prominent feature formation. Perturbations integrin-matrix signaling by administration either colchicine or anti-αvβ3 antibody resulted dissolution tubular association with increased apoptosis. The activation αvβ3 integrin induced expression anti-apoptotic gene bcl-2 and conferred resistance to proapoptotic effect TGF-β1. In contrast stable networks formed HUVEC, bovine aortic (BAEC) exhibited more dynamic process formation spontaneous involution. inhibition BAEC transfection prevented involution network. TGF-β1 present within system mediated inducing death. Indeed, blockade neutralizing antibodies reduced preserved As observed HUVEC networks, during were also dependent on survival-promoting effects matrix-integrin interactions. This study suggests structure determined balance vs. signals engagement cytokine receptors milieu. J. Cell. Physiol. 178:359–370, 1999. © 1999 Wiley-Liss, Inc.
