摘要: BACKGROUND Loss of mucosal integrity is associated with intestinal hyperpermeability, which may be inhibited by glutamine. The nature this effect unknown. glutamine on protein kinase C (PKC)-mediated hyperpermeability in HT-29Cl.19A cells was studied. METHODS Confluent monolayers HT-29C1.19A were cultured permeable filters and mounted Ussing chambers for permeability studies. Apical to basolateral transepithelial intact horseradish peroxidase (HRP) determined. Phorbol-12,13-dibutyrate (PDB) used activate PKC-mediated the (0.6 mmol/L) RESULTS Two hours PDB stimulation increased HRP flux, reaching five times control values after 4 hours. Bilateral exposure reduced PDB-induced (37%). Preincubation 2 before showed an earlier greater (3 hours, 43%; 50%). This bilateral mimicked separate apical exposure. Basolateral alone had no effect. CONCLUSIONS Glutamine rapidly cells. dependency suggests that more effective when delivered enteral route.
