Identification and genetic effects of a novel polymorphism in the distal promoter region of porcine leptin gene.

摘要: Leptin is a hormone expressed mainly in white adipose tissue. It acts on satiety centers the hypothalamus, and by binding to particular receptor OB-Rb, plays crucial role controlling appetite, body weight, fat amount total energy balance. In this study, novel single nucleotide polymorphism (SNP), i.e., SNP G-2863A, was identified distal promoter region of porcine leptin gene. Totally 780 individuals Duroc, Yorkshire, Laiwu, Lulai Black breeds, Landrace × Yorkshire crossbred were genotyped PCR-SSCP approach. The effects G-2863A mRNA serum protein levels as well driving transcription examined. results showed that, except for Duroc pigs, three genotypes GG, GA AA detected. Allele G occurred with higher frequency western breeds whereas lower Laiwu pigs. Statistical analysis indicated that there consistent trends toward animals genotype GG than or backfat thickness both circulating level half-sibling activity allele significantly A (P < 0.01) reporter gene expression. These suggest potential DNA marker has regulatory transcription.
