MeCP2 Is a Transcriptional Repressor with Abundant Binding Sites in Genomic Chromatin

摘要: MeCP2 is an abundant mammalian protein that binds to methylated CpG. We have found native and recombinant repress transcription in vitro from promoters but do not nonmethylated promoters. Repression nonlinearly dependent on the local density of methylation, becoming significant at bulk vertebrate genomic DNA. Transient transfection using fusions with GAL4 DNA binding domain identified a region capable long-range repression vivo. Moreover, able displace histone H1 preassembled chromatin contains methyl-CpG. These properties, together abundance high frequency its 2 bp site, suggest role as global transcriptional repressor genomes.
