摘要: Skeletal muscles of patients with Duchenne muscular dystrophy (DMD) and mdx mice lack dystrophin are more susceptible to contraction-induced injury than control muscles. Our purpose was develop an assay based on the high susceptibility limb in for use evaluating therapeutic interventions. The involved two stretches maximally activated tibialis anterior (TA) situ. Stretches 40% strain relative muscle fiber length were initiated from plateau isometric contractions. magnitude damage assessed one minute later by deficit force. At all ages (2-19 months), force deficits four- seven-fold higher compared mice. For muscles, unrelated age, whereas increased dramatically after 8 months age. increase older did not parallel similar adverse effects mass or production. situ stretch protocol TA provides a valuable investigations mechanisms dystrophic test interventions reversing DMD.
