摘要: The thymus is the central lymphoid organ providing a unique and essential microenvironment for T-cell precursor development into mature functionally competent T-lymphocytes. Thus, it important to develop strategies enhancing thymic regeneration from involution induced by variety of clinical treatments conditions. Hepatocyte growth factor (HGF) promotes proliferation in cell types. We have used stem cell-based HGF gene therapy enhance acute involution. HGF-overexpressing human adipose tissue-derived mesenchymal cells (HGF-hATMSCs) were generated liposomal transfection with pMEX expression vector, constructed inserting gene. Significantly increased these was confirmed reverse transcription-polymerase chain reaction an enzyme-linked immunosorbent assay. produced HGF-hATMSCs enhanced mouse epithelial line interleukin-7 vitro. also examined effect on rats Significant increases size weight, as well number thymocytes (especially, early thymocyte progenitors), seen HGF-hATMSCs-treated compared saline-treated control animals. A stimulatory has therefore been shown, highlighting value treating
