Effects of surrounding sequence on the suppression of nonsense codons

摘要: Using a lacI-Z fusion system, we have determined the efficiency of suppression nonsense codons in I gene Escherichia coli by assaying beta-galactosidase activity. We examined four amber suppressors acting on 42 different (UAG) at known positions gene, and UAG suppressor 14 UGA codons. The largest effects were found with supE (Su2), which displayed efficiencies that varied over 35-fold range, suppressor, 170-fold variation efficiency. Certain sites so poorly suppressed (less than 0.2%) they not originally detected as mutations. Suppression can be correlated sequence 3' side codon being suppressed, many cases first base side.
