Cloning and expression of a transcription factor activator protein-1 member identified from the swimming crab Portunus trituberculatus

摘要: Transcription activator proteins are regulatory that bind to the promoter regions of target genes. protein-1 (AP-1) regulates numerous genes related immune system, apoptosis, and proliferation. In this study, full-length cDNA AP-1 from Portunus trituberculatus (PtAP-1) was identified by expressed sequence tag analysis cDNA-end rapid amplification. The gene is 1183 bp encodes a 256-amino acid protein with predicted molecular mass isoelectric point 28.96 kDa 8.90, respectively. PtAP-1 showed highest expression level in gonad tissue lowest blood, hemocyte, muscle, hepatopancreas, gill, during first 6 h low-salinity stimulation (10%). Additionally, we observed steady decreases mRNA but at 12 h, initially upregulated, followed significant decrease until restoration baseline levels 48 h. Vibrio alginolyticus challenge resulted upregulation 6 h, which maintained high for From 48 72 h, levels, although they remained significantly higher than those detected baseline. These results suggested involved response osmoregulation crustaceans.
