Dual-Agent Photodynamic Therapy with Optical Clearing Eradicates Pigmented Melanoma in Preclinical Tumor Models.

摘要: Treatment using light-activated photosensitizers (photodynamic therapy, PDT) has shown limited efficacy in pigmented melanoma, mainly due to the poor penetration of light this tissue. Here, an optical clearing agent (OCA) was applied topically a cutaneous melanoma model mice shortly before PDT increase effective treatment depth by reducing scattering. This used together with cellular and vascular-PDT, or combination both. The effect on tumor growth measured longitudinal ultrasound/photoacoustic imaging vivo immunohistology after sacrifice. In separate dorsal window chamber model, angiographic coherence tomography (OCT) generated 3D tissue microvascular images, enabling direct assessment response. had minimal therapeutic control, non-pigmented melanomas but statistically significant (p < 0.05) lesions for both single- dual-photosensitizer regimes. latter enabled full-depth eradication tissue, demonstrated absence S100 Ki67 immunostaining. These studies are first demonstrate complete response immunocompromised vivo, quantitative volume thickness, confirmed (immuno) histological analyses, as controls clarify critical role melanin results indicate potential OCA-enhanced lesions, including melanoma.
