摘要: Transgenic maize plants expressing dsRNA targeting western corn rootworm (WCR, Diabrotica virgifera virgifera) DvSSJ1 mRNA, a Drosophila snakeskin (ssk) ortholog, show insecticidal activity and significant plant protection from WCR damage. The gene encodes membrane protein associated with the smooth sepate junction (SSJ) which is required for intestinal barrier function. To understand active RNA form that leads to mortality of larvae by interference (RNAi), we characterized transgenic transcripts mRNA. expression silencing cassette results in full-length transcript 901 nucleotides containing 210 bp inverted fragment gene, formation double-stranded (dsRNA) siRNAs plants. Our artificial diet-feeding study indicates dsRNAs greater than or equal approximately 60 base-pairs (bp) are activity. Impact specificity mRNA on activities was also evaluated diet bioassay, showed single nucleotide mutation can have impact abolish against WCR. These provide insights as functional forms plant-delivered feeding damage information contributing risk assessment dsRNA.
