Evaluation of different promoters driving the GFP reporter gene and selected target tissues for particle bombardment of Dendrobium Sonia 17

摘要: Three different morphological callus types, identified as type A, B and C, tips of in vitro inflorescences were used target tissues for genetic transformation. Five DNA plasmids carrying a synthetic green fluorescent protein (gfp) gene driven by promoters, CaMV 35S, HBT, Ubi1 tested the transformation Dendrobium Sonia 17. 35S-sgfp-TYG-nos (p35S) with 35S promoter showed highest GFP transient expression rate, while HBT promoters relatively lower rate all tested. The number GFP-expressing cells was observed on day 2 post-bombardment, declined gradually over course next weeks. Type A found to be best potential
