摘要: Abstract The objectives of this work were to evaluate the direct effects diallysulfide (DAS) and diallyldisulfide (DADS), two major organosulfur compounds garlic oil, on mitochondrial function integrity, by using isolated mouse liver mitochondria in a cell-free system. DADS produced concentration-dependent swelling over range 125–1000 μM, while DAS was ineffective. Swelling experiments performed with de-energized or energized showed similar maximal amplitudes. Cyclosporin A (1 μM), ethylene glycol-bis(2-aminoethylether)-N,N,N′,N′-tetraacetic acid (EGTA, 1 mM) ineffective inhibiting DADS-induced swelling. minor (12%) decrease membrane protein thiols, but did not induce clustering proteins. Incubation (but DAS) an increase oxidation rate 2′,7′ dichlorofluorescein diacetate (DCFH-DA), together depletion reduced glutathione (GSH) increased lipid peroxidation. dissipation potential, cytochrome c release. DADS-dependent effects, including swelling, DCFH-DA oxidation, peroxidation loss inhibited antioxidants iron chelators.
