摘要: A novel cDNA clone termed E16 which codes for an integral membrane protein of 241 amino acids with six transmembrane domains was isolated from peripheral blood lymphocytes. The is 4000 base pairs in length and exhibits unusually long 3'-untranslated region about 3000 nucleotides. Its expression at the mRNA level closely linked to cellular activation division. In all myeloid lymphoid cells, as well primary lymphocytes blood, transcripts are rapidly induced degraded after stimulation. This pattern unusual resembles more kinetic seen protooncogenes lymphokines T cell system. isolation made possible by a approach especially designed selectively cDNAs exhibit such kinetic. It based on combination differential screening subtracted library subsequent hybridization resulting phages short oligonucleotide (5'-TAAATAAA-TAAATA-3'). complementary trimer rapid degradation signal (AUUUA) present single or reiterated motif many short-lived transcripts.
